I don't get this perspective at all. Dean is obviously attractive, as was in evidence the last episode with the other waitress. Is the show expected to have women fawn over Dean in every episode? Is every waitress supposed to trip over herself to get with Dean? When characters don't find Dean attractive, it's comical because it subverts the expected. Just like when Crowley uses nicknames for Sam that imply he's stupid. The audience knows it's not true, so it becomes a joke. Same with Dean -- the audience knows how handsome Dean Winchester is. I don't think any of it is TPTB saying that Sam is dumb or Dean is unattractive. They're just jokes.
