In the scene here, they are actually seeing "suspicious activity" (a skeevy-looking dude leading away a young woman) outside the goth-ish Club Spider... which is the vampire they are looking for who was giving the new "drug" (vampire blood) to unknowing young women. Dean has on his furrowed brown, "hmmm that doesn't look at all good" face and Sam has on his "uh oh, I think this shit's going down now" face. I think the brightness was correct on your image AwesomO, they've brightened things back up again since S6, but S3 was really, really bright and colorful so sometimes when I see a really colorful image I just automatically go to thinking it's S3.
